Description

FOXO4-DRI is the compound behind one of the most widely shared images in longevity research: aged mice with visibly restored fur and improved physical function. That study, published in the journal Cell in 2017 by Peter de Keizer's laboratory, is real and rigorous. The mechanism is called senolysis, selectively killing senescent cells, which are damaged cells that stop dividing but refuse to die and instead sit in tissue releasing inflammatory signals. FOXO4-DRI disrupts a protein interaction those cells depend on for survival. Everything demonstrated for it was demonstrated in mice. There is no completed human trial and no confirmed first-in-human study. The photographs are genuine, and the conclusion people draw from them about human use is not supported.

What Are The Claims

Claims made for this compound: Selectively clears senescent cells via FOXO4-p53 disruption (mouse); Restored some fitness measures in aged mice; Proposed anti-aging and healthspan benefits

Evidence

The senolytic mouse study that made this famous was a single striking result. No human has been studied. Self-experimentation with an unreplicated senolytic is a meaningful unknown, not a calculated risk.

Evidence Strength

Animal/preclinical only

Caveats

NO human trials; a single landmark mouse paper; injectable human use is entirely experimental.
