Metformin (off-label longevity use)
Description
Metformin activates AMPK, a master cellular energy sensor that mimics some of the metabolic effects of caloric restriction, and large observational studies in diabetics have noted lower rates of cancer and all-cause mortality compared to non-diabetics on other medications. This drove the ongoing TAME (Targeting Aging with Metformin) trial interest and its adoption by some longevity-focused physicians for non-diabetic patients, despite the human longevity benefit remaining unproven in healthy populations. Importantly, metformin can blunt some of the muscle-building and cardiovascular adaptations from exercise when taken around training, which is why many longevity physicians have patients time doses away from workouts. This is a prescription medication — it requires physician oversight, regular kidney function monitoring, and is not something to self-source.

How it Feels

Metformin may also blunt some adaptations to exercise training. Use only under physician supervision.
